A groundbreaking clinical trial has offered fresh hope to a three-year-old boy suffering from chemotherapy-resistant liver cancer, with the child reportedly achieving remission following an innovative treatment. The young patient, who has not been named, was diagnosed with metastatic hepatoblastoma, a rare and aggressive form of liver cancer that primarily affects children. This type of tumour often develops in children under the age of three and is considered the most prevalent form of childhood liver cancer, as noted by the National Cancer Institute.
Prior to the experimental treatment, the boy had undergone three courses of chemotherapy and several surgical interventions, including a complete removal of the primary liver tumour and two procedures to address lung metastases. Unfortunately, despite these attempts, the cancer swiftly returned, leaving healthcare professionals with limited options.

Faced with the child’s deteriorating condition, the medical team enrolled him in the CARE study, an ongoing clinical trial aimed at assessing the effectiveness of CAR T-cell therapy for young patients battling recurrent solid tumours. This therapy employs genetically modified T-cells to target and eliminate cancer cells, and has shown promise in various clinical contexts.
During the trial, the child received two infusions of GPC3-CAR T-cells, which were specifically designed to target the glypican-3 protein prevalent in hepatoblastoma cells. Remarkably, after the first infusion, researchers noted a partial response. Following the second infusion and a lymphodepletion phase—intended to enhance the efficacy of the treatment—imaging revealed that the metastatic disease had resolved, leaving only residual scarring.

Lead researcher Dr. David Steffin, who is the associate chief of cellular therapy and bone marrow transplant at Texas Children’s Hospital and an associate professor at Baylor College of Medicine, expressed optimism about the results. He stated, “This case demonstrates that a durable complete remission in a patient with a chemotherapy-resistant solid tumour can be accomplished entirely in the outpatient setting without systemic toxic effects.”
